About the Spectroscopic Structure Elucidation certification
An expert-level certification for chemists who already know organic chemistry and basic instrumental analysis and now need to determine structures they have never seen before. You will learn to run the full elucidation workflow the way it is done in pharmaceutical, natural-product, process and forensic laboratories. It starts with the molecular formula: degrees of unsaturation, exact masses and ppm errors from high-resolution mass spectrometry, isotope patterns for chlorine, bromine and sulfur, carbon counting from the M+1 peak, the nitrogen rule, the choice between EI, CI, ESI, APCI and MALDI, and the fragmentation logic of alpha-cleavage, the McLafferty rearrangement and neutral losses. Infrared spectroscopy is treated quantitatively — diagnostic regions with exact ranges, the carbonyl ladder and the reasons behind it (conjugation, ring strain, induction, resonance, hydrogen bonding) — together with UV-Vis chromophores and the Woodward–Fieser rules. The heart of the course is NMR: proton chemical shift and anisotropy, integration, first-order coupling and Pascal triangle patterns, geminal, vicinal (Karplus), aromatic and alkene coupling constants, complex multiplets, diastereotopic protons and exchange; carbon-13 shift ranges, symmetry counting, DEPT editing, the NOE, dynamic NMR and fluorine and phosphorus couplings; and the 2D toolkit — COSY, TOCSY, HSQC, HMBC, NOESY and ROESY — read cross peak by cross peak and assembled into fragments. The final module solves complete unknowns that combine every technique, then closes with absolute configuration by X-ray crystallography (Flack parameter), electronic and vibrational circular dichroism, and the Mosher method, plus a systematic workflow and the reporting standards journals expect. Every lesson has a hand-built animated figure drawn to exact spectroscopic scale, worked examples with real numbers, common pitfalls and self-checks. This credential certifies that you can take raw spectra of an unknown and defend a complete structure.
